Sharmila was born and raised in Pune in a middle-class farming family. Her father, who holds a BSc in Agriculture, later chose to become a banker. Sharmila is the first in her family to enter the entertainment industry. She graduated from art school with a BFA in Applied Arts.

Sharmila began her career in 2010 with the show Swapnanchya Palikadale on Star Pravah. Her major breakthrough came in 2011 when she portrayed ‘Rupali’ in one of the most popular Marathi shows, Pudhcha Paaul, also on Star Pravah.
Initially, Sharmila used her birth name, Sharmila Shinde, for her screen name. However, in 2017, she started using her father’s name as her last name on screen, becoming Sharmila Rajaram. In a press interview with a reputable newspaper, she explained that she changed her last name because she believes an artist should not be judged by their name but by their skills, and that an artist has no religion, caste, or creed. Despite this, she switched back to her previous name in 2019. This name change led to some confusion, with some media referring to her as Sharmila Shinde and others as Sharmila Rajaram.
Sharmila is well-known for her work in the Marathi show Mazhya Navryachi Bayko, where she starred alongside Anita Date-Kelkar, Rasika Sunil, and Abhijit Khandkekar. She has an upcoming movie in 2024 titled Lifeline, featuring Madhav Abhyankar and Ashok Saraf.
